#1 - Salmonellosis in calves without intestinal lesions

Abstract in English:

Salmonellosis is a known cause of enteric disorders in calves. However, cases in the septicemic form may not present enteric lesions, which may lead the veterinary practitioner to not suspect salmonellosis, compromising the diagnosis. The current study describes the epidemiological, clinical, pathological and immunohistochemical aspects of septicemic salmonellosis in calves without enteric lesions. The protocols involving bovine material submitted to the Pathology Laboratory (LAP) of the “Faculdade de Medicina Veterinária e Zootecnia” (FAMEZ) of the “Universidade Federal do Mato Grosso do Sul” (UFMS) from January 1995 to July 2018 were studied. Cases confirmed or suggestive of septicemic salmonellosis in calves without enteric manifestations were selected. Fragments of the liver, lung, and spleen embedded in paraffin were submitted to immunohistochemistry (IHC). Only cases in which there was positive marking on the IHC or culture isolation of Salmonella were included in this study. Of a total of 5,550 cattle examined in the period, ten presented septicemic salmonellosis without enteric lesions. Clinical signs included mucosal pallor, apathy, hyperthermia, and dyspnea. Only three calves presented diarrhea, and two were found dead before clinical changes were observed. The most common necropsy findings were hepatosplenomegaly; yellow, orange or brown discolored livers; pale mucous membranes; inflated and sometimes red lungs; fibrin or fluid within body cavities; and gallbladder filled with inspissated bile. Jaundice was observed in three calves that had a concomitant infection with Anaplasma sp. Microscopically, paratyphoid hepatic nodules and interstitial pneumonia were the most frequent manifestations, followed by thrombosis and bacterial colonies in the spleen, lung, liver, and brain. A strong positive marking was observed in IHC, predominantly in the lung and to a lesser extent in the liver. Polymerase chain reaction (PCR) indicated the Dublin serotype as the causative agent in the samples of the four calves submitted to this procedure. In calves, the septicemic form was the major cause of death due to salmonellosis. Septicemic salmonellosis was usually not accompanied by diarrhea. The clinical signs of septicemia are nonspecific and of little assistance in the diagnosis. IHC has been shown to be efficient in the detection of the agent, mainly in the lung and especially in situations where it is not possible to perform bacterial culture.

A salmonelose é uma causa conhecida de distúrbios entéricos em bezerros. Porém, casos na forma septicêmica podem não apresentar manifestação entérica, o que leva o médico veterinário a não suspeitar de salmonelose, comprometendo o diagnóstico. Este estudo descreve os aspectos epidemiológicos, clínicos, patológicos e imuno-histoquímicos da salmonelose septicêmica em bezerros sem lesões entéricas. O estudo foi realizado a partir dos protocolos referentes a materiais de bovinos enviados para diagnóstico ao Laboratório de Anatomia Patológica (LAP) da Faculdade de Medicina Veterinária e Zootecnia (FAMEZ) da Universidade Federal de Mato Grosso do Sul (UFMS) de janeiro de 1995 a julho de 2018. Foram selecionados os casos de bezerros confirmados ou sugestivos de salmonelose septicêmica sem lesões entéricas. Fragmentos de fígado, pulmão e baço embebidos em parafina foram submetidos ao exame de imuno-histoquímica (IHQ). Somente foram incluídos neste estudo casos em que houve marcação positiva na IHQ ou isolamento da bactéria em cultura. De um total de 5.550 bovinos examinados no período, dez apresentaram salmonelose septicêmica sem lesão entérica. Os sinais clínicos incluíram palidez de mucosas, apatia, hipertermia e dispneia. Apenas três bezerros apresentaram diarreia e dois foram encontrados mortos sem terem sido observadas alterações clínicas. Os achados mais frequentes de necropsia foram hepatoesplenomegalia, fígado amarelado, alaranjado ou acastanhado, palidez de mucosas, pulmões inflados e, por vezes, vermelhos, fibrina ou líquido nas cavidades do organismo e vesícula biliar repleta de bile grumosa. Icterícia foi observada em três bezerros que apresentavam infecção concomitante por Anaplasma sp. Microscopicamente, os nódulos paratifoides hepáticos e pneumonia intersticial foram as manifestações mais encontradas, seguidas por trombose e colônias bacterianas no baço, pulmão, fígado e encéfalo. Na IHQ, marcação fortemente positiva foi observada, predominantemente, no pulmão e, em menor intensidade, no fígado. A técnica de reação em cadeia de polimerase (PCR) tipificou o sorotipo Dublin como agente etiológico nas amostras dos quatro bezerros submetidos a este procedimento. Em bezerros, a forma septicêmica foi a principal responsável pelas mortes por salmonelose. Na maioria das vezes essa forma não estava acompanhada por diarreia. Os sinais clínicos da forma septicêmica são inespecíficos e de pouco auxílio no direcionamento do diagnóstico. A IHQ mostrou-se eficiente na detecção do agente principalmente no pulmão e especialmente nas situações em que não é possível a realização da cultura bacteriana.


#2 - Intestinal lesions in pigs slaughtered in Rio Grande do Sul, Brazil

Rio Grande do Sul stands out in the national swine industry due to high production rates, based on an intensive breeding system, which leads to the occurrence of enteric diseases. This study was performed to diagnose and determine the frequency of causes of bowel condemnation in the slaughter line. The samples of intestine and mesenteric lymph nodes were collected for histological and immunohistochemical analysis of a pig slaughterhouse located in the Vale do Taquari area, in Rio Grande do Sul (RS), which has Federal Meat Inspection Service (SIF). The collection occurred during 18 days in the summer (January and February) and 19 days during winter (June, July and August) of 2016. The pigs were from 55 different counties from of seven microregions of RS (microregion of Passo Fundo, Soledade, Guaporé, Santa Cruz do Sul, Lajeado-Estrela, Montenegro and São Jerônimo). Out of 2,523 condemned intestines in the summer and 2,747 in the winter, 216 and 199 were evaluated, respectively. The three main diagnoses were chronic serositis (43.1%), ascaridiosis (21.4%), and porcine proliferative enteropathy (20%), followed by cases of plug caseous in ileocecal valve (5.0%), intestinal abscess (1.0%), pneumatosis (1.0%), circovirosis (0.7%), oesophagostomiasis (0.7%), salmonellosis (0.7%) and a case of intussusception (0.2%). Chronic serositis was diagnosed in 82/216 (38%) in summer and 97/199 (48.7%) in winter, ascaridiosis had a higher occurrence in the summer with 64/216 (29.6%) compared to the winter when it was observed 25/199 (12.6%). Porcine proliferative enteropathy was diagnosed in 37/216 (17.1%) in summer and 46/199 (23.1%) in winter.

O Rio Grande do Sul se destaca na suinocultura nacional pelos altos índices de produção, baseada em sistema de criação intensivo, fato que propicia a ocorrência de doenças entéricas. Este estudo foi realizado para diagnosticar e determinar a frequência das causas de condenação de intestinos na linha de abate. As amostras de intestino e linfonodos mesentéricos foram colhidas para análise histológica e imuno-histoquímica, de um frigorífico de suínos localizado na Região do Vale do Taquari, no Rio Grande do Sul (RS), que conta com Serviço de Inspeção Federal (SIF). A colheita ocorreu durante 18 dias nos meses de verão (janeiro e fevereiro) e 19 dias nos meses de inverno (junho, julho e agosto) de 2016. Os suínos eram provenientes de 55 municípios diferentes, que fazem parte de sete microrregiões do RS (microrregião de Passo Fundo, Soledade, Guaporé, Santa Cruz do Sul, Lajeado-Estrela, Montenegro e São Jerônimo). Dos 2.523 intestinos condenados no verão e 2.747 no inverno, 216 e 199 foram avaliados, respectivamente. Os três principais diagnósticos foram serosite crônica (43,1%), ascaridiose (21,4%) e enteropatia proliferativa suína (20%), seguidos por casos de tampão caseoso em válvula ileocecal (5,0%), abscesso intestinal (1,0%), pneumatose (1,0%), circovirose (0,7%), esofagostomíase (0,7%), salmonelose (0,7%) e um caso de intussuscepção (0,2%). A serosite crônica foi diagnosticada em 82/216 (38%) no verão e 97/199 (48,7%) no inverno, ascaridiose teve maior ocorrência no verão com 64/216 (29,6%) comparado com o inverno em que observou-se 25/199 (12,6%). #3 - Intestinal lesions in pigs affected with postweaning multisystemic wasting syndrome, p.313-318

Pigs originated from 12 farms in southern Brazil and were selected on the basis of clinical signs and/or gross lesions suggestive of enteric disorder. Lymphohistiocytic infiltrates of varying intensity were associated with anti-porcine circovirus type 2 (anti-PCV2) immunostaining (IS) in samples of intestines and mesenteric lymph nodes from all pigs. Although most findings were similar to those described in PCV2-associated enteritis, anti-PCV2 IS in association with depletion of the goblet cell mucin stores (24 pigs), diffuse ileal villous atrophy and fusion (18 pigs), and dilatation of the lymphatic vessels (11 pigs) combined or not with lymphangitis were also observed. PCV2 antigen was immunohistochemically demonstrated in the cytoplasm and nuclei from intralesional epithelial cells, histiocytes, and endothelial-like cells in intestinal tissues. Together these findings imply an association with PCV2. The presence of co-infections by Lawsonia intracellularis, Brachyspira spp., Mycobacterium spp., Salmonella spp., rotavirus, parvovirus, coronavirus and enteric calicivirus with PCV2 in the intestinal lesions was investigated.

#4 - Atypical mycobacteria isolated from intestinal lesions of Bovine Oesophagostomosis

Abstract in English:

It was found that 90% of 1 to 9 year old cattle from southwestem Brazil had nodular intestinal lesions of Oesophagostomosis in the final portion of the ileum and in the cecum. The number of nodules varied from 1 to 53 and their diameter from l to 19 mm. The cut surface of the larger nodules showed a dry yellowish caseous material, and some times fragments of Oesophagostomum radiatum larvae could be observed. Bacteriological examination of 400 samples taken from the intestinal wall with and without lesions resulted in isolation of 42 (5,2%) cultures of atypical mycobateria. Thirty cultures originated (71,4%) from nodular material and 12 (28,6%) from the portion without lesion. Twenty three (54,8%) cultures belonged to the Runyon group IV and 19 (45,2%) to group II and III. There were 4 cultures of Mycobacterium scrofulaceum, 1 of the M. terrae-complex and 8 of M. intracellulare. Fifteen (78,9%) of these cultures were isolated from the nodular lesions and only 4 (21,0%) from the part without nodules. The present results suggest that the nodular lesion caused by Oesophagostomun radiatum larvae is a predisposing factor for the penetration and multiplication of atypical mycobateria in the intestinal wall. Consequently inespecific allergic reactions to tuberculinization may develop.

Na região sudeste do Brasil foram encontrados nódulos de esofagostomose na mucosa intestinal da porção final do íleo e do ceco em 90% dos bovinos, variando a faixa etária de 1 a 9 anos de idade. O número de nódulos variou de 1 a 53, e o tamanho destes de 1 a 19 mm de diâmetro, tendo a maioria de 2 a 3 mm. Ao corte, os nódulos maiores continham massa caseosa, ressecada de coloração branco-amarelada, às vezes esverdeada. Em alguns foi observada a presença de larva de Oesophagostomum radiatum. A cultura de 400 materiais com nódulos e outros 400 sem, permitiram o isolamento de 42 (5,2%) culturas de micobactérias, sendo 30 (71,4%) oriundas de materiais com nódulos e 12 (28,6%) dos materiais sem nódulos. A identificação das culturas revelou que 23 (54,8%) pertenciam ao grupo IV de Runyon, consideradas saprófitas e 19 (45,2%) se enquadraram nos grupos II e III, consideradas facultativamente patogênicos, sendo 4 culturas Mycobacterium scrofulaceum, 1 do complexo M. terrae e 8 de M. intracellulare. Destas 19 culturas, 15 (78,9%) foram isoladas da porção da mucosa com nódulos e apenas 4 (21,0%) da mucosa sem nódulo. O presente resultado sugere que as larvas de O. radiatum predispõem a penetração e multiplicação de micobactérias atípicas nos nódulos da esofagostomose e, consequentemente, estas provocam sensibilização alérgica inespecífica observada na tuberculização.
